RMVX plays MIDIs through DirectSound, so SW Synth doesn't affect playback.

I know but how i can do the ME in-game?

You go to an event and go through the options and select "PLAY ME"
What is "PLAY ME"? I din't find it!

I tried and it's does not work even. They are MIDIS. Some unknown reason it can't play MIDI files.
Either it can't work...there's a problem that the sound can't play, i have ordered the key of Rpg Maker VX, the problem is same as RPG Maker 2000 did if i installed.

I'd really like to get rid of LockeZ. His play style is way too unpredictable. He's always like this too. If he ran a country, he'd just kill and imprison people at random until crime stopped.

You probably want music to be under BGM, not ME. Regardless, whichever folder you put it in (BGM or ME), that's the event command it'll show up under.
Is it just this file that can't play, or all midi files, or all music files of any format?
Does it fail to work in the real game, in test play, or both?
Does it even show up in the list of songs to play?
