In Japanese animation and drawing, the single fang is an indicator of uneven teeth. It's considered 'cute' and often seen in 'moe' girls (Kyouko from Madoka, for example).

BTW, I do find it rather amusing that she goes around saying that she's not a witch, she's not a witch -- yada-yada-yada -- but judging by what she's wearing -- HOW ARE THESE IDIOTS NOT SEEING THIS!?!?!?

(I know; anime people are usually pretty slow and used to the whole people walking around in strange clothing sort of thing.)
